Clinical significance
- Anti-double stranded DNA antibody (Anti-ds DNA) is one of a group of autoantibodies called antinuclear antibodies (ANA). Normally, antibodies protect against infection, but autoantibodies are produced when a person's immune system fails to adequately distinguish between "self" and "non-self "
- Anti-ds DNA specifically targets the genetic material (DNA) found in the nucleus of a cell, hence the name "anti-dsDNA." The anti-dsDNA test identifies the presence of these autoantibodies in the blood
- While anti-dsDNA may be present at a low level with a number of disorders, it is primarily associated with SLE. Anti-dsDNA test is used to help diagnose lupus (systemic lupus erythematosus) in a person who has a positive result on a test for antinuclear antibody (ANA) and has clinical signs and symptoms that suggest lupus
- A negative anti-dsDNA does not rule out lupus. An anti-dsDNA test may be ordered along with a test for anti-Sm (Smith antibody), another antinuclear autoantibody associated with lupus, to help establish a diagnosis
- Antibody to DNA is followed by the appearance of circulating DNA antigen and subsequently the formation of immune complexes deposited in the kidney glomeruli initiating kidney damage
- It helps in prediction of progress of the disease assisting in therapeutic decision making
- Can show disease remission with reduction in the anti DNA antibodies and normalization of C3/C4 levels
- Sudden increase in SLE disease activity are generally preceded by a rise in anti ds-DNA levels, followed by a steep drop at the height of the disease
When to get tested
- When there is s/of lupus ( Muscle pain, Arthritis-like pain in one or more joints , malar rash, Low-grade fever, Persistent fatigue, weakness, Inflammation and damage to organs and tissues, including the kidneys, lungs, heart, lining of the heart, central nervous system, and blood vessels)
- When ANA test is positive
